detection of nestin on cytospin-preparation of the neuroblastoma cell line SK-N-LO by ABC-immunocytochemical staining with alkaline phosphatase.
Using Blocking Solution leads to a strong reduction of unwanted two-dimensional background. After correct staining of cytoplasmatical nestin also the haematoxylin counterstain of the cell nuclei is not problematical any longer for the interpretation of results.

detection of CaMKII in lysate from heart and brain tissues of C57Bl/6 mice.
Monoclonal Anti CaMKII was used as primary antibody and a polyclonal HRP conugate as secondary antibody.
Only the combination of Blocking Solution and LowCross-Buffer® (as dilution buffer for primary and secondary antibodies) leads to a clear result with visible bands.

detection of osteocalcin in osteoblast culture.
A fresh osteoblast culture shows, on the first day, no or very weak expression of the extracellular matrix protein osteocalcin.
Using Blocking Solution the situation is presented correctly (shown left side, first picture). Standard blocking with BSA leads to a false positive result (right side).
With increasing duration of cell culture, extracellular matrix and osteocalcin is building up.
The increase can be tracked correctly by using CANDOR The Blocking Solution. The cultures are stained intensively.
